I see you have already created a thread to most of your questions pertaining to sponsorship/application that you have post to this thread.
Most likely you will get a quick reply from the forum, but to answer a few of your questions, yes start gathering up all require documents, you don't have to mail your package from Columbia, you can definetly bring her portion of the application with you to Columbia and have her complete it with you, it will be your choice to submit your completed package either from Columbia or Canada.
Visiting Canada - yes she can visit while your application is in process - see CIC website or threads in the forum pertaining to spouse visiting while application is in process - consider yourself lucky if she gets approved !!
medical - it's best to do it upfront and submit with completed application to CIC - see CIC website or threads in the forum pertaining to sponsored person medical - you will need to follow the correct guide for each country pertaining to medical - panel physicians only !!
Supporting documents - yes all must be submitted with your complete package to CIC - there are excellent threads on the forum pertaining to supporting documents and how to prepare your package.
Mailing the package - the best way to mail your package (if in Canada) is through Canada post express services that way you can track your package to CIC - keep your tracking slip so you will know when your package arrived and who signed for it - it will probably be Mahmood lol (I'm sure some of the other know what I mean lol - but that's not important, as long as its signed by CIC
The Forum is the best place to assists you with your immigration process to sponsor your spouse - your in the right place, but this thread your in is basically discussions, experiences, process and status of all October 2015 Outland applications who are now going through the process (we already submitted our applications )
so pretty much we come here to support each other and discuss our experiences .
Whichever month and year you submit your application I'm more than sure they will have a thread (similar to this one) set up so you to can discuss the process of your submitted application.
