With respect to sponsorship, you want to try to demonstrate as much as possible to the VO that you have funds readily available in Nigeria to support your studies. I am guessing you have already received your LOA. So a few things to consider, have you paid any of your tuition at all, if not, then ask your uncle in-law to pay for at least the first year of study. Once that is done, you will get an official receipt from the school, and that will show commitment. If you uncle in-law has assisted with that already, all you need to show in your husband's Naira account is the amount you need for upkeep for at least a year. That is a minimum of about 12,000CAD (just to be on a safe side). Then you husband will provide letter of udertaking to support your studies while you in Canada, and I think with that you should be fine. Do not give the VO any hint that you are depending on someone in the US for your sponsorship.
Who has assured you of this job placement on completion of your studies, your current organisation, if yes, kindly include the letter. This is a good motivation to the VO that you have interest in returning to Nigeria on completion of your studies.

No. Remember you already show an intention to return to your home country based on the letter assuring you of a position when you get back, if that is the case, the VO would be wondering why does she want to take her family with her. That is contradicting yourself and enough grounds to show dual intent and hence refusal of your application. As a matter of fact your husband and kids staying behind shows strong family ties that will motivate you to return on completion of your studies. In summary it is best you get your permit first, before applying for them.
