Hiw many reference letters are ideal? One is enough?
 
One for each work experience you're claiming points from under CRS.

Is it important that reference letters list all duties and responsibilities for all jobs, even for those that you are not getting any points?

I have a few that only confirms that I was there and what my pay was.

You question is already answered above by Ufa. You only need reference letters listed with roles & responsibilities for only those jobs that you are claiming points towards CRS.

If you list your spouse's work experience in EE profile then yes, you'll have to provide reference letters for it. But, as it doesn't get you any points, you can skip it and add it in personal history after ITA.

Is that after getting ITA or while creating EE profile? Am already in the pool awaiting ITA with CRS 431

Ok, if you list your spouse's work experience while creating your express entry profile it will not give you any extra points but you would have to provide reference letter for it.
If you want to avoid providing experience letters for your spouse do not mention any of her/his work experience in your Express Entry profile. Just list her/his work experience in personal history when you fill in your after ITA application.
