im the sponsor..my spouse and I had lived together for 2 years before getting married.she went back home to her home country february 2011.
in her application questionaire ask if I send her money..we put NO. because since we live together i never send her money and we dont have any proof of transfer if we said yes.

are they going to put a red flag on that because i dont send her money?

after i submitted our application(feb2011)I send money to her once..if i continue sending her money, can we still use the receipt of money transfer as proof of genuine and continuing relationship in case they will call her for interview?

If money is sent does that show the relationship is genuine? Will it help or no?

Sending money to a spouse is one possible indication that the relationship is genuine. If you do send them money, certainly say so in the application and provide proof. But if you don't, don't worry about it. It is just one type of evidence - it is not necessary. In many situations, neither spouse needs money from the other, so it doesn't even apply to these people.
If you send money after sending in the application, keep the proof. You can take it to the interview as additional proof of contact and support.

Also keep in mind that sending money is not always seen as good. Make sure you have lots of other proof as well. One Nigerian man I know sent money to his Canadian wife, and the visa officer accused him of paying her to marry him.

In your case it should bd fine. The sponsor sending money to the applicant rarely looks suspicious, especially if the applicant needs money because of the conditions of his home country.
