I have sponsored my wife from the USA who had overstayed her visa. Now finally after 5 months our application got approved in the USA by the NY immigration office. They are asking us to submit our passports in order to get the Visa to Canada. I wanted to know at what point do they issue us a PR card and once we have the PR card does my wife need a visa to enter USA.

After you get the PR visa in your passport, you go land in Canada and at that point give them an address to get the PR card mailed to. It's supposed to take 4-6 weeks to get it. Being PR of Canada means absolutely nothing to the US. If your wife needs a visa for the US now, she will need one in the future until she goes for Canadian citizenship.

You will have to deal with the US embassy in Canada to apply for her visa. Since she overstayed in the US in the past, she may have problems with that.
