Hi I am working on Open work permit in Manitoba for last 6 months, however my spouse is on closed work permit in Ontario,so it will effect my PNP process, though my score is more than 700 after shows my spouse status.
Please advise me what I supposed to do, If I don't include her than what happened next ??

Yes it will affect, you will lose 100 points for her work and if one of you attended a college/university you'll lose more 100 points - 200 points in total. Why wouldn't you include her if you said you have more than 700 points after shows your spouse status?

You have to include your spouse on your MPNP application.

If you are on spousal open work permit, you have to wait till you complete one year of work experience in Manitoba.
