Did you check YES or NO in the column where the question was asked? You have already explained the refusal, so that should be fine. You should attach a short explanation to the new form stating that you made a mistake, and give a short recap of the explanation of the denial.
It is misrepresentation, and the PR visa could well be denied because of this, so send in an amended form.

Definitely send in a new form with the correct information and an explanation. If the application is rejected because of this, you would probably win on appeal, because you did include the details in your application. The judge might then find that your intent was not to deceive. However, you do not want to go to appeal - it takes a lot of time, and you should really have a lawyer for it, so it will be expensive.
The best course of action is to just submit a new version of the form, with the correct answer and an explanation of the circumstances of the refusal, and an explanation of why you answered differently before.

The visa officer is already going to have questions, because you answered 'no', but then explained that you had been refused a visa. Sending in a corrected form might postpone the process a bit. Not correcting this might postpone the process more.
