In case employer is not giving Reference letter with Job duties, can I take a reference letter from my colleague on a plain paper with his contact details, who is at same or level than me. Taking letter from a Manager / Supervisor would be ideal but it could be damage the internal growth prospects.

where you should declare the reason for not getting LOR and explain your main duties, working hours,designations details etc etc.etc to support your claim, you should provide your contract/appointment letter, Certificate of employment (you can take this from your HR, without any issue), salary slips.
this is the best option you can opt for, in my opinion.
