I have mailed to CIO for the status of my application and got reply that my applicaiton is under review and will contact in due course.

What do other seniors say about my situation? As my file received on 27th Sep and just 2 hours ago I read that the Acedamic Transcript should be in a seal cover,and I have put all Notarised trascript with my application, what will happen next (again, the file will come back)? What should I do next?

all u can do as of now is wait .. if they email back a rejection(the probability of which is high) u will have to wait for the pplication packet to come back and then resubmit.. if u can arrange a new admission packet with the university sealed transcripts then u may submit it as soon as u receive the rejection email

else if in case it gets accepted.. then u r lucky...

About transcripts: Can the people who got PER's so far confirm that you ALL sent them sealed into envelopes from Uni's? Or Legalized copies would do...?

Does this mean that other assessment and eligibility review such as education, background check , ... will be done in VO?!?!?!?!
In that case next step is not medical check and it will take more time for them to review and assess our documents in VO

I assume so, the reason being the local visa office is supposed to be specialist for that country. It knows the value of the applicant's education certificates, can call up provided references during the standard office hours in that country, and so on. At least, that was how it worked under the old system. It's possible more of the burden has been moved away from the VO to CIO but it's impossible to know as we're the first ones through this new system.
