As part as the in Canada Sponsorship application you have a section of medical check ups(It is in the check list provided).
I paid something like 200$ if I'm not mistaken and went to to a doctor office in Vancouver.
They tested blood & urine took an x ray and quickly took my weight and height and also inquired about my general health.
End of day I got three slips as prove of payment and sent them with the application as requested.
They have my information in their database.

If you really want a driver license here are my recommended steps:
1.Learn the theory material from the book and be prepared.In addition go online and take the test provided there,its very helpful.
You should be ready so if the official approve you for taking the test you will do it on the spot and pass(very easy test took me 10 minutes but I read the book more than 4 times just to be sure)
2.Print:
*The email you got from CIC
*Screenshot of you on line status with your name in it("we received your application on...status") and the same one with your spouse status on line so they can see your name with the word "permanent residency" near it
3.Bring your passport and your previous driver license(if applicable)
4.The most important thing-Bring your husband! They react differently when talking to Canadians(I brought my wife) with a Canadian accent
5.Go to the Burrard branch
6.After the official(usually young and confused) talking to you will get confused he will go to his manager(And if not demand it).
The manager will decide if to approve you for testing or not.I was refused by the first one and approved few days later with a different one so it all depends on the manager and her/his mood
