I got my work permit extension approved and got a letter from IRCC confirming the same. Once I receive work permit, can I travel to India and get passport stamped with visa in India? My father is admitted to hospital and I may need to travel to India urgently.

You need to be holding a valid TRV to return to Canada. You can apply for the TRV from outside of Canada.
The bigger issue is TRV processing times can be quite long so if you leave Canada you may have difficulty returning for a long time.
