I have calculated my score through CRS points calculator and received 382 points. Please let me know if my score will increase after education evaluation and if so by how much. I have a Masters degree in Pharmacy.
Having ECA will definitely increase you CRS .... it will give you education point as well as skill transf. points ...so go ahead and do it as soon as you can ...as the score you currently have will not give you invite (ITA) ...
ECA is still being processed. I have a masters and bachelors degree in pharmacy. I have claimed points for ielts, age, education, experience. Is it necessary to do ECA for spouse also if she is a dependent applying along with me?

No. I'm afraid you misunderstand the procedure. You will earn your 126 points for education ONLY if you get your credentials evaluated. Without the evaluation, you won't be awarded any points (and won't be admitted into the pool at all). When filling in the Express Entry profile in the education section, you will be asked if you have your education evaluated by an authorized organization (and you'll be asked for the eval. case number). If you select "no" to the evaluation, your education won't count at all.
To sum up, evaluation won't add anything to 126; it's the condition on which you will be awarded points for your education at all.
Your overall CRS score will increase if you retake IELTS (getting at least 7 for each section except Listening and 8 for Listening, the more the better). This will add more than 40 points (you can use the same tool to calculate how many exactly).
It's even better if your wife also takes IELTS (if she hasn't already).
