You have to mention your spouse details no matter where she is based.
As you are the principal applicant, you can request Buffalo. Your wife is just a dependent in your case.
If called for an interview, she should be there definitely. But that is during the final stages of the application process.
We applied under NOC 0631, the documents reached CIC on July 18th. We didn't hear from CIC yet, also anticipating some news.
