If my husband requests to extend his stay as a visitor while waiting for outland PR, are we able to take a trip outside Canada (say down south for one week) during the time of his visa extension? Or would it cancel the extension of his visa?

Once he leaves Canada, his visitor status ends. On return he has to satisfy the VO that he is admissible for entry.

The moment your husband leaves Canada, he will lose his visitors status including his extension. He would have to convince the border agent to let him back in again. And if he managed to get back in, he would only be granted 6 month max stay. He would have to re-apply for another extension again if he wishes to stay longer.
