study permit to PR

My situation is, I am a U.S. Citizen who is leaving for Canada on a study permit soon. As for applying for PR, do I have to wait until I am there for a year or can I apply to the Buffalo as soon as I get there?

As a US citizen, you can apply to Buffalo at any time, that is if you qualify.

Yes, I am getting married when I get to Canada, and will be applying for PR Family Class, I just wasn't sure if I could do that as soon as I got up there. But thank you!

One more question is, with me already being in Canada, can I still apply outside Canada? I understand I am obligated to travel if I need an interview, but I just wanted to make sure I didn't have to apply in Canada because I will be up there. I want to make it the quickest process which most research has pointed to applying outside.

Yes, you can apply via the outside route even though you are living in Canada.
