If he is travelling to Canada to visit his sister, will that count as essential? If he does get his visitor visa and they get to travel tgt and quarantine tgt for 14 days will that still allow him to accompany her?

He will need to get authorization to travel as well as a TRV. The process is here under extended family member travel.

I got admission for MBA in Canada now and I want to apply for the study visa. I have been refused US visiting visa before but after then I applied for UK visity visa without making reference to the US refusal and I got the visa. Now, do I need to mention the US visa refusal in my Canada Study visa to be applied for shortly?

Yes - you must mention the US refusal. Canada and the US share immigration information. If you fail to mention the US refusal, you can expect your study permit to be refused and you may also end up with a 5 year ban from Canada for misrepresentation (lying in your application).
