My brother who is a Canadian national can sponsor or send invitation letter to 02 families at the same time? Or is it better to finish one case and then start a new one? Please guide

There is no such thing as sponsoring people for a visitor visa. Your brother is inviting them. Your brother is not supporting them and both families have to show their own funds, travel history, ties to home country and reasons to travel. So he can write multiple invitation letters.
