Hi, my wife studied her full time MBA program in Toronto from September 2019 and I got my Spouse Open Work Permit last week and I have just received a job offer working as audit manager at an accounting firm.

Am I eligible to apply for Express Entry? If yes, how long do I need to wait before I get PR?

You are eligible to apply for permanent residency if you receive enough CRS points (around 460-470), there are many different things they assess to see if you have enough points. Generally you need to have good English, some work experience, well educated, extra points for age, permanent resident family member etc

You can apply for Express Entry at any time.

Note that you cannot claim the 50 job offer points with the job offer you have now. To claim the 50 points, your employer would need to obtain an approved LMIA.

Many thanks for this reminder and I think this is the most tricky one. Can I ask my employer to help get the approved LMIA after I start work at that company? Then I can claim the bonus points?

Yes - if your employer can obtain an approved LMIA, then you can claim the 50 points.
 
Even with completion of one year in the company requires getting a LMIA for claiming the 50 points?

Depends on what kind of work permit you hold. If you hold an open work permit like a PGWP or SOWP, then yes, your employer needs to obtain an approved LMIA for you to qualify for the 50 points.
