I am currently living in Saskatchewan on OWP and received a job offer letter. The offer letter mentioned a 3-month probationary period. My question is can I claim points based on the job offer letter ? Because while creating the application it's written that High skilled employment offer from a Saskatchewan employer gives 30 points. My NOC belongs to teer 2 so my question is can I claim point for that ? Or the job needs to be a permanent one because no such thing mentioned while submit the interest application form.

So the employer is approved by SINP and you have received Job Approval Letter from SINP for the job? Your employer would have filled out the SINP forms. If you do and meet all other requirements if the ISW-Employment offer category then you can count the points.

When you submit the expression interest form no such thing is written that your employer needs to be approved by SINP plus you need JAL to claim that point. It's just written "High skilled employment offer from a Saskatchewan employer "

You are misreading the chart you posted. This is for the Employer Offer subcategory only which requires JAL. Check out the requirements for Employer Offer.
