Hi,
I hope so someone can answer my questions
1. you have a Post graduate work permit and it going to expire match 2015, but got an job offer example contract job, can the person extend their Post graduate wokr permit?
2. Would I need a LMO when I apply.
I know there some situations where a LMo is not needed. So someone please Explain.
Thank you.

You need an LMO/LMIA to extend your work permit past the expiry of your PGWP. You are not LMO/LMIA exempt. Information on who is exempt can be found here:

http://www.cic.gc.ca/english/work/apply-who-permit.asp

Technically, you cannot extend a PGWP, but you can apply to extend your work work permit with an LMIA (LMIA is a new term to replace LMO, but the idea is the same).

2. Would I need a LMIA when I apply.
Yes. However, if your employer has already applied for the LMIA and you are still waiting, and your work permit is about to expire (within 2 weeks), you can apply with the LMIA pending - you would have to apply BEFORE your PGWP expires, and you would have to provide proof of the LMIA application (e.g. copy of the application, proof of delivery, correspondence from ESDC...)

LMO exemptions are for situations such as those covered by free trade agreements (e.g. NAFTA), see the link that scylla posted -

If you have a year of experience you can apply for permanent residency under Canada experience class, and then apply for bridging work permit..

Apply for PR and if you meet the criteria, you can apply for a bridging open work permit (BOWP). Easiest is CEC, which requires 1 year work experience in Canada.

You could try looking at the PNP programs to see if you are eligible in that category.

LMIO getting tougher, strict conditions on employers to which positions they can apply LMIO for.

PR is your best bet

a) what province are you in?
b) what kind of medical assistant? Clinical or administrative? Do you know your NOC?

You might be eligible for PNP, if your employer is willing to help. You would have to apply within 2 years of graduating...

http://www.ontarioimmigration.ca/OI/en/pnp/OI_PNPSTUDENTS_JOB.html

At some point in the process, I'm not entirely clear when, you become eligible for a work permit.

Alternatively, if your employer applies for and receives an LMIA, you could be eligible under the Arranged Employment Stream of the Federal Skilled Workers program..

http://www.cic.gc.ca/english/immigrate/skilled/apply-who-instructions.asp?expand=arranged#arranged

But it all depends on how soon you would complete the 1 year work experience requirement of the CEC class to determine which option makes the most sense..

If you reached 1500+ hours of work and if you jobs falls under NOC 0,A,B.
If would call CIC or go to the nearest Immigration office and ask them, If you could apply for PR under CEC as you completed the required number of hours.

Let me know what they say.
