In both case you need to make your express entry profile. So make your profile and see how much point you have. Ontario provincial nominee program is now closed. So wait until they open

Are you still working?

If you are and have LMIA you can get 600 points.

If I apply for express entry profile first. Will I be able to apply for OINP when it opens. If Ontario nominates me through OINP, can I update my express entry profile which I had previously created?

If you are referring to the Human Capital Priorities Stream, the only way you are going to receive a PT is to have an Express Entry profile at the first place.

Did you not read the previous replies? You need to have an express entry profile open before Ontario will nominate you. If/when they do nominate you, 600 points will be added on to your CRS, no need for you to do anything else.

If you did masters from UfT, Univ of Ottawa, or UWO in specific program, you don't need express entry profile to apply master stream OINP. It's a separate process.
