Does anyone know the process of getting an expedited/same day work permit at the Niagara or any international border?

Whether that's possible or not really depends on your circumstances. You need to provide full details if you want someone to answer with any accuracy.
I applied for PGWP in November 2015 and it was refused because I did't attach the letter from school stating that all requirements had been met by me and then I applied for restoration of status and work permit (Paper application) on March 26, 2016 but I forgot to pay the work permit fee and I realized about this mistake when I called CIC to inquire about my application status. I paid the fee and mailed the receipt to CIC.scylla said:
The receipt was received by CIC on July 4, 2016. The online status says that are working on applications received until March 12, 2016. I am concerned if they will consider fee receipt sent by me.
And If they don't consider my fee receipt and sends back the application, can I go to Niagara Border and get my work permit.
I am not able to work and my employer wants me to start ASAP.
Meanwhile I had filed for my PR under express entry in March 2016.
I hope this clarifies my situation.
If you leave this will be considered as a completely new entry... And yes they probably may not be willing to grant you your work permit, even though nothing is really preventing them from doing so. (Talking from experience)
Considering you have a PR in progress, I'd suggest you wait. It shouldn't be too long now.

The alternative could hurt your application for PR, get you in trouble, etc. or maybe work perfectly fine (if the WP can be asked at a POE). There is no way to know. It's up to the agent at the border.
Having been in this situation, I tried Niagara twice... to be rejected for no valid reasons and be treated like sh*t. I then requested a pre-approval letter for the WP, when to the US for a few days, and tried my luck at Pearson. It worked, but those were extremely painful and stressful days.
