No you cannot. Quebec do not follow Express Entry. There is a high chance that your express entry application will be rejected, if you have work & residency in Quebec. Your chances of getting a PR via Express Entry from Quebec will be 50-50.

You can but would have to show solid proof of intent to reside outside of Quebec.

The best way is to apply for the PEQ : https://www.immigration-quebec.gouv.qc.ca/en/immigrate-settle/temporary-workers/stay-quebec/application-csq/workers-peq/index.html

And when you have your CSQ you apply to the PR for Quebec.
