My wife is filling out her PR application from the Netherlands. She's on the background section where you list out information about your family. She is an only child currently as her older sister went missing when she was a child and her whereabouts are unknown. Does she still need to list them or it's better to omit?
 
It's better not to omit anything. Why should you? As if it may hinder your application somehow. Most likely it will have no effect on the application. Hiding something will, if found, make her inadmissible on the grounds of misrepresentation.

Absolutely do NOT omit. Put the information she has, and if something is missing, include a short letter of explanation about why. If information is included, nothing to worry about.

Would this letter just be uploaded under supporting documents?
 
Under supporting documents, there is category for Other. If we have multiple sections where we need to explain something, do we need a letter for each? I ask because you can only select a category once.

Don't know, you'll have to figure it out.
 
Isn’t there a category for letters of explanation. You should probably submit under that.
