No. But you would have a job offer with an employer to be eligible.
Any periods of self-employment, unauthorized work, or periods of employment during which you were engaged in full-time study (ex: work experience gained on a co-op permit) will not be included when calculating the period of qualifying work experience in Manitoba. https://immigratemanitoba.com/immigrate-to-manitoba/skilled-workers/swm/eligibility/

Thank you,
I don't understand. So they would hire me as TFW and I have to work for that employer 6 months so I can apply like it says there or I can apply with a job offer?

You need to be working for an employer for at least 6 months and the employer has offered you full time employment. It is both.
Ongoing Manitoba employment means that you possess a valid work permit and a Manitoba company has offered you a long-term, full-time job after you have completed at least six months (continuous) full-time employment with that company as a temporary foreign worker or international student working graduate.
