My employer owns several shops in the same province and he is saying that he may need to assign me to a different location, not stated on our LMIA although it's within the same province. Is this alright, would it affect our work permit application? Do we need to do anything?

You need to work in the location on your LMIA. If your work permit is being processed for another location then you would need to apply for a new work permit.

I have closed work permit for 3 years, Employer office is in Alberta and Ontario. In LMIA my company location was mentioned as Ontario office.

In my work permit , below conditions are mentioned :-

Conditions:
1. MUST LEAVE CANADA BY 202******
2. NOT AUTHORIZED TO WORK IN ANY OCCUPATION OTHER THAN STATED.
3. NOT AUTHORIZED TO WORK FOR ANY EMPLOYER OTHER THAN STATED.
4. NOT VALID FOR EMPLOYMENT IN BUSINESSES RELATED TO THE SEX TRADE SUCH AS STRIP CLUBS, MASSAGE PARLOURS OR ESCORT SERVICES.

Can I work from my Alberta's office location without any issue ?

Where will your payroll run?
Should be fine for now

If you apply for PR in future, you can expect some questions around this or even PFL in some cases - So make sure to document everything

Payroll will run from Alberta

Its saying BARRIE
Barrie is in Ontario
So ideally you should reside in the Province where you are paying Taxes on your payroll

It is a very common practice that some remote job folks do end up staying in other locations but rest assured expect IRCC to question you at some point. There is a good amount of risk
