If i am on the work visa and I have valid job offer letter and I am working with a company.
do I still need to get that LMIA approved to get scores in Express entry?
 
My company is US based and they have a subsidiary here in Canada.
My job offer of manager level?

Is it valid ?
or what do mean by valid?
 
In order to get the 50 or 200 points for having a valid job offer (and since you do not have LMIA) you must:

1 - Have an employer-specific work permit (closed work permit)
2 - Work for that employer for one year.
3 - Have a job offer that states that you will be working for such employer for a year after becoming a permanent resident.

Otherwise you do not have a valid job offer and you cannot claim those points.

Suppose after getting PR with LMIA and refuse to work then what happened?

Once you're PR you can safely quit your job even if you claimed a valid job offer.

i have just got LMIA with fulltime permanent job offer.

is it mandatory to have work permit with LMIA to get PR approval in EE?
