Shall I need to send a letter from my job & contract?

how can I prove that I am in the NOC of 38?

you don't. they just look at your list of duties and your NOC code and decide whether you qualify or not.

Then in round two, they do the same check again but this time with the proof that you send from your employer, etc.

Where do they look at the list of duties? Which form is this? Or is this through the resume? I didn't see that in the list of forms to be submitted though.

 Bank draft for the required amount made payable to the Receiver General for Canada
 Application for Permanent Residence in Canada (IMM 0008)
 Schedule 1: Background/Declaration – Principal Applicant
 Schedule 1: Background/Declaration – Spouse
 Schedule 3: Economic Classes – Federal Skilled Worker
 Additional Family Information (IMM 5406) – Principal Applicant
 Additional Family Information (IMM 5406) – Spouse
 Photocopy of passport’s bio-data page
 Photocopy of Identity Card
 2 self-addressed mailing labels in English (or one in your country's official language)
