Do I need to submit a letter of enrollment/registration for study permit extension even if applied for a new course at a different college? Also, what does returning students exactly mean?

Returning students is just for Registrars office business. They keep you in shortlist for accepting you in the program ( This sometimes doesn't mean you are registered yet in the program depending on the college/university).
If you need to extend your study permit you need a official acceptance letter without any admission acceptance conditions. If you apply with an enrollment letter and a returning student document there are fair chances that your application will be refused.

It's not required but I highly recommend you do. I've seen posts where students have been denied extension due to not submitted letter of enrolment.
