I had 35 hr/week work experience as RA in south korea for more than 3 years. I got reference letter from my prof. for it. Is RA countable as 35 hr/week job?
Is research assistant (RA, NOC 4012) countable as 35 hr/week job?

I think so. As long as your letter matches the main duties (note some TA duties may be needed) and paid, you are good.
Note in another discussion someone pointed out "4012 will be harder for RA because most duties in 4012 are TA related. " So solely RA may be problematic.
