First, yes, you have to list him, even if you don't know his address in Canada. Not listing him can get you banned if CIC finds out.
As far as it effecting your visitor's visa... I don't have any experience to answer this question, other than to say, yes I would imagine it would negatively effect your chances for a visa. CIC may think you're really planning to join your brother illegally in Canada.

Write "unknown."
Then attach a short letter stating you are unaware of your brother's current location and relationship status. I had to write one about my mother (of all people) when I applied for my PR visa. It basically said, "I have not seen or spoken to my mother since 2005. I am unaware of her current address or martial status. I did ask my father and he is also unaware." It must be reasonable. If you can ask your parents and they can provide information without much problem, then you should provide the information.

You need to make all reasonable measures to provide an address. If you cannot, then 'unknown' is your best option - unless your brother truly is missing. If you report him missing inside Canada, the authorities will look for him. If he's working illegally, this won't end well.
