Your situation is a little different than other applicants who are residing in the United States.
You are in-canada applicant, so if there is an interview scheduled, you will be required to travel to the united states to attend the interview, therefore, yes, you will have to apply for a visiting visa to the USA, and if your visa gets refused, you will have to inform the visa office "Buffalo" as soon as possible providing a proof of your claim "the refusal letter from the american embassy in canada".
In such a situation, the visa office will do one of the following scenarios:
Transfer your application to the visa office serves your country of nationality, and inform you officially about that decision.
Or conduct the interview via phone.
Or ask for additional documents which was the reason of the lack of information led to the interview decision.
Or Waive the interview without asking for any documents, if the interview was just a random selection for quality assurance in your application.
There will be no refusal of your permanent residence application only based on the fact that you are not able to attend the interview because of the above mentioned reason, as long as you notify them about the visa refusal in the proper time providing the official supporting documents of your claim.
The selection of any of above scenarios will be decided by the visa office not you, so all you have to do is to notify them, and they will decide which scenario to follow with your case.
