40.6K is in my account, spouse account 5K, Mother account 10K.
My wife has applied for OWP and coming with me.

So your suggestion is to pay first year tution fees to university?
All money belongs to my mother and spouse transfer to my account? Isn't any problem sudden transaction to my account? Can't my mother and wife sponsor me?
Why should the visa officer believe that you can pay for your tuition and living expenses if the money is not in your bank account?

I don't believe that it adds much value in the eyes of the VO if you paid your tuition fees. You need to show financial capability. Also, as others said... Keep maximum funds in your account only. Not a consolidation of accounts.

Big transfer are fine if they can be justified by a valid source of income like salary, selling of assets, etc. However, transferring it from your family's account into yours will create red flags as VO in Canada don't consider family funds as yours even though culturally in Asian countries this is very much acceptable.

You can either pay 1 years or 1 term fees whatever your college requires. But I would recommend paying some fees atleast.

Noted with thanks. I got your point. Another thing, if I pay one year tution fees to university, should I clarify the source of that money during application or not required. I am asking because I want to borrow this money from my aunt and will return her after got approval. So that, my existing money will not deduct from my account. I think you get my point.

You probably can but the problem I see here is, you have already shown your statement before and they will have a record of this. When you re-apply with tuition fees paid, they might pick on this as the balance has not changed. But no harm trying as I dont see you have many options at this point.
