Fresh application for PR - Employment Basis - from Buffallo

Hello,
I am currently in US on H1 and am interested in making a fresh application for PR from Buffallo NY.
I am very new to this and would appreciate tips from Guru's.
First thing I am trying to understand is do I apply under :

Federal Skilled Workers
For Provincial Nominees
Quebec Skilled Workers

Also, what know of timeframe am I looking at from making an application to confirmation of PR.

Start reading at www.cic.gc.ca and take the assessment, either here or on the CIC site to see if you qualify. Then take a look at the PNP sites to see if you would qualify there http://www.cic.gc.ca/english/skilled/provnom/index.html
