I am Canadian Permanent Resident and my sister is in Canada who just got the ITA for Express Entry. She has put my information while creating profile, giving her 15 points.
Now, the situation is I got my Green Card approved and will be moving to USA soon.
Would that affect my sister’s PR application ?
Her points =444, cut-off on the draw= 417

Yes if you move before she gets PR she needs to remove you from her application. Sibling points are awarded because family in Canada has been shown to lead to a more successful settlement. Unfortunate but misrepresentation also has very serious consequences.
 
Would add that if you are sponsoring a parent you must advise IRCC that you are leaving Canada and you no longer will qualify to sponsor your parents.

My brother is PR in Canada and got married 15 days back.
Now he is sponsoring his wife who is in India and simultaneously my wife who is on work permit in Canada will apply for express entry.

As my brother will apply for wife sponsorship, can we at the same time claim 15 points in our express entry profile?

Yes. As long as he is living in Canada.
 
So that means he can apply for wife sponsorship along with helping us to claim his points for express entry simultaneously
