After filling the express entry profile, in how many days acknowledgement number comes?

You mean reference EE No.?

Generally immediately.

You don't get any "Acknowledgement number" after creating your EE profile. As soon as you "Transmit" your application, you will receive your EE profile number. If you have not got it, then it means that you haven't transmitted your profile properly.
 
so after getting EE profile number (eligibility check) any other number comes from the CIC acknowleding the profile?

You will get an email with a pdf attachment that will have your 'Job Seeker validation code' generally a 4 digit code, that will be required for your JobBank account creation/activation.

Should no be more than an hour at max.

Mine took a few minutes say 15-20, don't remember exactly.

I have got EE number and registered with job bank for NOC 4021 (Computer Science Tutor) for my wife. Overall she got 364 points.

Could you please tel us how much is the possibility of getting ITA either on NOC code demand and final CRS score?

The ITA is issued solely based on your CRS score. Don't want to disappoint you but 364 points has very little chance of getting ITA in 2016.

1) Try to appear in IELTS again. If you and/or your spouse can reach CLB9 then it will boost your CRS to above 400 levels
2) Another option would be to try for PNP of various Canadian provinces. If you can get a nomination then it adds 600 points to CRS score meaning guaranteed ITA in the next draw
