Review "valid employment" definition by CIC, you will see that it requires LMIA to get points, if you don't have LMIA, forget it.

I was working there as a supervisor while I was in school and after my studies, I got promoted to assistant manager. Do I still require LMIA?

“Valid job offer” definition is clearly showed on CiC website, you need to have Lima to get 50 points unless your job is Lima exemption. Check it out.
