I'm thinking of starting a business that would require speaking my wife's native language as there would need to be correspondence and customer relations back in her home country. Are there any restrictions to sending out an LMO for your own wife?

No, but the business doesn't exist, and you can't specify a language other than French or English, so a LMO is extremely unlikely to be approved.

I would apply for one after the business exists and French and English would be a requirement since it would be based in quebec but I also need someone who speaks her native language as I will be dealing with hospitals in her native country who likely don't speak English or French!

Anyone else?

You can state that in your LMO application that the native language is required, but at the same time you have to convince then she has to be inside Canada to undertake the business. You have to meet other requirements as well.
My questions would be, why not sponsor her if your purpose is to bring her to Canada, or invite her on Business Visa for both visiting and working?

She is already in Canada and her work permit is about to expire. PR takes about 2 years to process from the country she's from.
