Can I apply for a work permit from inside Canada?

Good day everyone.. I am having a hard time understanding the legal process to get my work permit..
So I hope someone can help me.. this is my current situation:
I am from Brazil, I am currently in Canada (holding a multiple-entree visitor visa, that is valid until 2017)..
While visiting, I got a offer to work as a Live-In Caregiver. The employers will start my LMO process.

My question is: if I get a positive LMO, can I apply from inside Canada for the work visa or must I leave the country to do so?
If I need to leave the country, is it necessary for me to go back to Brazil for that or can I apply in the USA (I also have a multiple-entree visa for USA as well)?

If you get your LMO, go south across the border, turn around, go back to enter canada, produce LMO, job offer letter and passport, pay $150 to the visa officer (make sure by checking official sources that the amount is correct). Go to work.
