Or, do you have to work for a certain amount of time first before you can apply for PR?

I assume the only way to apply for a skilled worker PR is through Express Entry, right?

If you have a job offer in Canada supported with a positive LMIA, then it will fetch you additional 600 CRS points for your express entry profile and you can receive ITA in the next round of invitation.
 
Also it is a must that you are eligible for either FSW or CEC before you create your express entry profile.

The answer to your question in the subject line is no.

As the poster above mentioned, you must still meet the minimum criteria for one of the immigration programs, even if you have an LMIA-approved jo offer.

All programs require a minimum of 12 months skilled work experience and passing a language test.

And yes, the Federal Skilled Worker program requires you to apply through Express Entry.

No. Eligibility for PR and LMIA approved job offer are two different things. You need to first check for your Eligibility under following 3 programs:

Federal Skilled Worker Program,
Canadian Experience Class, and
Federal Skilled Trades Program.

You need to be eligible under at-least 1 of these programs before you can claim the 600 additional points from LMIA approved job offer.
