Applying for PR while on TRV or Super Visa

if someone is in Canada on TRV or Super Visa, can he / she apply for Permanent Residence while in country ?

Yes - you can apply for Family sponsorship while in Canada. However applying won't let them stay in Canada until the processing of the application is complete. They will still have to return home at the end of their visit - even if their PR application hasn't been fully processed yet.

my older son is a permanent resident, waiting on his citizenship application to be approved,
will he be able to apply for family sponsorship to sponsor his Mother AND young SIster (9 years old) on the same application , provided he qualifies ...?

Yes - he will be able to include his mother and sister in the same application provided he has the income required to qualify and meets the other requirements. Note that CIC is not accepting parent sponsorship applications at this time. CIC will start accepting applications again early next year. Expect the processing of this application to take several years (4? 5? 6?).
