Best to call between 10am and 12noon. You may need to keep trying every 10 minutes till you connect. Although, I expect you to get a response to your mail on Monday.

Your call has a higher chance of connecting if you call within 10am and 12noon. I figured that out and got lucky twice. Mail was replied as well.

When did you get the mail? Did you send it through VFS originally? I'll advise you hold on till Monday and go back to VFS to check again.
