I have just graduated form my undergrad degree last year and am pursuing my masters in india now which would take two years. Would I be eligible to apply after I graduate from my masters ? Is it compulsory to work after masters and then apply? Or can I apply after I graduate from my masters ? I don't have prior experience in a job as of now .

you need at least 1 year of job experience to be eligible under Express entry Federal Skilled Worker. This program is for people who have a set number of skills that can be beneficial for the Canadian society. If you want to apply you need to first have one full year of experience under NOC O, A or B (google them)
