Job letter is as pharmacist and duties contain both pharmacist duties and assistant duties in description, will that work?

No. Your duties must match the NOC. You are a pharmacist trying to change your NOC to assistant. Not sure why you are trying to do that but would be misrepresentation.

Do you mean work as a pharmacy assistant in Canada? Did you read the Express Entry requirements for PR? If not, do so.

I have a masters degree in pharmacy but i have no experience working as a pharmacist so i was wondering can i work as a pharmacy assistant in my home country and apply for PR as pharmacy assistant

My points are 454 with one year of experience but i saw that pharmacy assistant is in demand so i was wondering if i can work as a pharmacy assistant and get a pr even though i have a pharmacist qualification?

Through EE or another program? Isn’t pharmacist in demand?

As long as you have one year of continuous, full time experience in that NO. Your score would not be high enough for current draws and the program may change before you are eligible so keep that in mind.
