A work permit and permanent residency are not directly related. Work permits are temporary in nature, permanent residency applications are permanent in nature.
You may QUALIFY for permanent residency based upon having worked in Canada for a sufficient period of time (two years as I recall, see the "CEC" information on the CIC website.
If you have an offer from your employer for permanent, full-time employment if you obtain permanent residency and that has been validated by HRSDC, you may qualify to file in the Federal Skilled Worker category ("arranged employment").
Information on both programs are on the CIC website. You may also qualify under a provincial nominee program, but I would suggest that you refer to your province's website for information on that.
