since you have an LMO, you can and should apply for a work permit. Having been refused a tourist visa may or may not have an impact depending upon the reasons of refusal. For e.g. if the VO thought you didn't show strong enough ties to return to your country and refused tourist visa, they would consider similar points when evaluating work permit. But all factors combined play role, the nature and type of job, your profile / qualification etc.
