I have atotal work experience of 2 years in last 10 years. but i am not not working for the last three years, will this affect my express entry application?

Nothing in the requirements mentions that your work has to be recent. As long as it's within the past 10 years, it should be ok.

I don't think so. Visa officers can't overrule the directions of CIC.
If CIC is fine with applicant who have work experience in the past 10 years, so will be visa officers.
There are many personal/medical reasons due to which a person might not be able to work for certain period of time.
CIC considers such reasons and so does visa officers.
