I have applied in NOC 2134 (chemical engineer) and in my reference letter, I have 5 work duties which match NOC 2134. My question is, How many work duties may seem to be satisfactory to the officer in charge?

It depends on how many job duties your selected NOC actually has. Usually, if your reference letter covers about 65-70% of the required duties, then you gonna be OK

Personally I would prefer 70-80% match.
