He needs to go through the LMIA process, including advertising the job for a month first to prove no Canadian can be hired for the role. Once the LMIA is approved, you then apply for a closed work permit. Make sure your nephew follows the advertising requirements. Advertising just in job bank is not enough. You will also want to make sure the job you are being offered in Canada is directly related to your past work experience / education.

Secondly, there have been some applications coming in, how will he be able justify to IRCC when applying for the LMIA?

I don't know how he jusifies the other applications when applying for the LMIA.
Expect that there will be significant scrutiny when it comes to your application to try to prevent exactly what you seem to be doing which is family members creating jobs to facilitate WP or immigration for family members not because there is an existing job that happened to be recently vacant. For example it sounds like your nephew created this position and someone had not recently left the position. How large is the company? Does your experience and education make you the ideal candidate over others? Does it make sense for you to have left your previous job to do whatever job is being offered? Would you have otherwise qualified for PR or a WP or is this job offer the only way you’d be able to secure a job offer? Your family is not the first family who has decided to create a job to hire a family member so IRCC is watching carefully because families abuse the system.
You will have to show why you are the ideal candidate for the job based on your previous work history and education. It can be very difficult to show that a Family members is the ideal candidate and a genuine job seeker not just trying to secure a permit or immigration.
You have to PROVE you are the best qualified to do the job , above anyone else that’s applied.
Thus the reason of the LMIA . If your relative is always going to hire you , then the process would be mute . Citizens / PR get priority over you . Did he already post for this position online PRIOR to you arriving ? Or did this position just become available since you’ve arrived?
Being related is irrelevant. IRCC is aware people “ create “ jobs for relatives just for immigration purposes
