my brother lives in Canada and I am attaching his citizenship card with my complete application. Should I need a covering letter from my brother where he may describe our relationship and provide possible support and guidance in future after landing in Canada? Is this letter could bring any benefit?

The letter won't carry any weight. You need his birth certificate, your birth certificate, showing the same parents. Proof of his status in Canada , and proof that he is residing in Canada, receipts, rent receipts, visa bills etc.

I also have a brother. I send his Canadian passport copy but was asked to submit further proof that he is in Canada and also establish proff that he is my brother. You need to send your birth certificates and his, marriage certificates, his latest credit card statements and any other proff that he is actually present in canada right now.

Yes, you have to have at least one common parent. So you can get 5 points based on your sister.
You need to show her and your Birth Certificate indicating that you have the same Father.
